摘要
目的建立喂养型代谢综合征(MS)大鼠模型,确定成模标准,评价成模情况,为药物疗效研究提供实验平台。方法 8周龄雄性SD大鼠随机分为正常组(10只)和实验组(50只),分别予正常饮食和高糖高脂高盐饮食喂养20周。每隔4周测量各组体质量、血压、空腹血糖,并分别于第4、12、20周末测量血脂(包括TC、TG、LDL-C、HDL-C)、空腹血清胰岛素等指标,计算稳态模型胰岛素抵抗指数(HOMA-IR)。结果实验组大鼠血压、体质量、空腹血糖、空腹胰岛素、HOMA-IR、TG、TC以及LDL-C水平均明显高于正常组(P均<0.05),HDL-C低于正常组(P<0.05),成模率达74%。结论喂养型大鼠模型符合MS的病理特征,操作简便,成模率高,可作为研究药物疗效的理想MS动物模型。
Objective To establish the rat models with metabolic syndrome( MS) by feeding,determine the standard of modeling,evaluate the modeling,and to provide experimental platform for the therapeutic study. Methods Sixty male SD rats aged 8 weeks were randomly divided into the normal group( n = 10) and the experimental group( n = 50),which were respectively fed a normal diet and high sugar,high fat,high salt diet for 20 weeks. The rats' body weight,blood pressure and fasting plasma glucose( FPG) were measured every 4 weeks. At the end of the 4th,12th,20th week,the blood lipids( TC,TG,LDL-C and HDL-C) and FINS were measured,and then HOMA-IR was calculated. Results The rats' blood pressure,body weight,FPG,FINS,HOMA-IR,TG,TC and LDL-C levels were significantly higher in the experimental group than in the normal group( all P < 0. 05),while HDL-C was lower than that of the normal group( P < 0. 05). The success rate of MS rat models was 74%. Conclusions This kind of rat models conforms to the pathological characteristic of MS,which can be easily operated and with high success rate. Therefore,it could be applied as an ideal MS animal model for the study of drug therapeutic effect.
