摘要
目的观察高迁移率族蛋白(HMGB1)对肿瘤血管生成的体外调节作用。方法使用HMGB1和血管内皮生长因子(VEGF)体外刺激人脐静脉内皮细胞(HUVECs)。通过细胞增殖试验、趋化试验、成管试验检测HUVECs的增殖能力、趋化能力、形成管腔的能力。结果 HMGB1可以在体外促进血管内皮细胞的增殖,趋化成管能力,且其促进作用呈剂量依赖关系(P均<0.05)。作为阳性对照的VEGF有明显促血管生成的作用。结论 HMGB1可以促进血管生成和肿瘤血道转移。
Objective To observe the regulatory effect of high mobility group box 1( HMGB1) on angiogenesis of human umbilical vein endothelial cells( HUVECs) in vitro. Methods We treated HUVECs with HMGB1 and vascular endothelial growth factor-C( VEGF-C) in vitro,and then we used CCK-8 assay,Transwell chamber model test,scratch and repair test and tube formation assay to detect the abilities of proliferation,chemotaxis,migration and tube formation. Results HMGB1 promoted the proliferation,migration and tube formation of HUVECs in a dose-dependent manner in vitro. As a positive control,VEGF-C significantly induced angiogenesis. Conclusion HMGB1 may promote the tumor angiogenesis and angiogenic metastasis,and this is expected to become the new direction in treatment of inhibiting metastasis of tumor.
