摘要 目的研究不同能量密度激光作用于姜黄素对人胃癌MGC-803的光动力学杀伤作用。方法 5μmol/L姜黄素与胃癌MGC-803细胞共同培养后,分别用不同能量密度9.6 J/cm2、19.4 J/cm2、40.2 J/cm2的波长为405 nm的激光照射,用MTT法检测胃癌MGC-803细胞株的增殖抑制率,流式细胞仪检测细胞的凋亡率及细胞线粒体膜电位、细胞内活性氧(ROS)及Ca2+的变化。结果随着激光剂量增大,细胞的抑制率及凋亡率呈上升趋势。能量密度为40.2 J/cm2的激光照射后,细胞株的增殖抑制率和凋亡率达到最大,分别达(44.93±3.61)%和(26.58±2.67)%,差异有统计学意义(P<0.05)。随激光剂量增大,细胞内线粒体膜电位显著降低,细胞内活性氧和Ca2+显著增加,能量密度增加为40.2 J/cm2时,细胞内线粒体膜电位降低至(66.2±5.8)%,细胞内活性氧升高至(115.7±5.8)%,细胞内Ca2+升高至(104.6+5.6)%,与其它组比较,差异有统计学意义(P<0.05)。结论以405 nm的激光照射,姜黄素对胃癌MGC-803细胞有杀伤作用,在一定范围内呈剂量依赖关系。 Objective To investigate photodynamic therapeutic effect of different energy density with curcumin on human gastric cancer MGC-803 cells. Methods 5 μmol / L curcumin co-cultured with gastric cancer MGC-803 cells was activated with 405 nm laser of different energy density 9.6 J / cm2,19.4 J / cm240.2 J / cm2. MTT was used to detect cell proliferation inhibition, and flow cytometry was used to detect cell apoptosis rate and mitochondrial membrane potential, intracellular reactive oxygen species(ROS) and Ca2+changes. Results With the increase of laser energy density, within a certain range, the inhibitory rate of cell proliferation and apoptosis rate increased. If energy density increased to 40.2J / cm2,the inhibitory rate of cell proliferation and cell apoptosis rate reached their peaks of(44.93±3.61)%and(26.58±2.67)% respectively(P<0.05). The intracellular mitochondrial membrane potential decreased significantly to(66.2±5.8)%,while intracellular ROS and Ca2+increased significantly to(115.7±5.8)% and(104.6±5.6)%, compared with other groups of different energy density. Conclusion Curcumin-mediated PDT with 405 nm laser has a synergistic cytotoxicity on gastric cancer MGC-803 cells in a dose-dependent manner within a certain range.
