miR-21促进SK-HEP-1肝癌细胞增殖和迁移的研究

摘要 目的研究促癌的miR-21在腹水来源的高转移人肝癌细胞SK-HEP-1的表达及对其生物学行为的影响。方法利用qRT-PCR检测miR-21在SK-HEP-1细胞中的表达。分别利用细胞活力和细胞毒性检测法(cellcountingkit-8,CCK-8法)、流式细胞仪及transwell小室评估miR-21对SK-HEP-1细胞增殖、凋亡及侵袭能力的影响。结果相比于肝正常组织,miR-21高表达于SK-HEP-1细胞(>5倍)。抑制其高表达后,SK-HEP-1细胞的增殖和迁移能力下降,但凋亡无明显变化。结论 miR-21高表达于SK-HEP-1细胞,并能促进其增殖及侵袭能力。抑制其表达可能成为治疗肝癌的新途径。 Objective To investigate the expression of miR-21 in SK-HEP-1 hepatoma cells and its effects on the tumor cell behaviors including cell proliferation,migration and apoptosis.Methods qRT-PCR assay was used to detect the expression of miR-21 in SK-HEP-1 cells and normal liver tissue.Cell counting kit-8(CCK-8),flow cytometery and transwell system were utilized to evaluate the proliferation,apoptosis and migration abilities respectively.Results miR-21 was highly up-regulated in SK-HEP-1 cells compared with normal liver tissue(>5 folds).miR-21 specific inhibitor significantly inhibited the proliferation and migration of SK-HEP-1 cells without significant change of its effect on apoptosis.Conclusions miR-21 can promote the proliferation and migration abilities of SK-HEP-1 cells.Knock-down of miR-21 expression by specific inhibitor may be a prospective therapy for hepatocellular carcinoma.
