丹皮酚对人结肠癌LoVo细胞增殖抑制机制的研究 被引量:4

摘要 目的研究丹皮酚对人结肠癌LoVo细胞增殖、凋亡的影响,探讨丹皮酚抗肿瘤的作用机制。方法用丹皮酚作用于体外培养的人结肠癌LoVo细胞,用MTT(四甲基偶氮唑蓝)比色法检测LoVo细胞的生长活性,光镜下观察细胞形态学改变,流式细胞仪测定细胞凋亡率以及Bcl-2蛋白表达水平。结果丹皮酚在0.047~1.504μmol/L剂量下对体外培养的结直肠癌LoVo细胞的增殖具有抑制作用,且呈明显的剂量、时间效应关系(P<0.05或P<0.01)。丹皮酚0.094~1.504μmol/L作用48h后,光镜可见LoVo细胞呈典型的凋亡形态学改变;流式细胞仪检测结果显示,细胞凋亡率升高,呈药物剂量依赖性,Bcl-2基因表达显著下调,与对照组相比,差异有统计学意义(P<0.01)。结论丹皮酚对人结肠癌LoVo细胞生长具有抑制作用,其机制可能与下调Bcl-2基因表达,诱导肿瘤细胞凋亡有关。 Objective To investigate the anti-tumor effect of paeonol ( Pae) on human colon carcinoma cell line LoVo and its molecular mechanisms. Method In vitro cultured human colon carcinoma LoVo cells were treated with different concentrations of paeonol.Cell proliferation was evaluated by MTT method and cell apoptosis was confirmed by morphology.Cell apoptosis rate and expressions rate of Bcl-2 were determined by flow cytometry. Results The proliferation of LoVo was inhibited significantly by paeonol in dosage(ranging from 0.047 to 1.504 μmol/L)and time dependant manners(P<0.05 or P<0.01). Typical apoptosis of LoVo cells was observed under microscope after incubation with paeonol for 48h. Cells apoptosis rate was significantly increased and Bcl-2 expression was down-regulated in a dosage dependent manner in paeonol group compared with those in the control group(P<0.01). Conclusions Paeonol can inhibit the proliferation of colon carcinoma LoVo cells. The anticancer mechanisms may be associated with down-regulation of Bcl-2 expression and induction of tumor cell apoptosis.
