淡紫拟青霉代谢产物生理活性物质分析和生物测定 被引量:11

摘要 淡紫拟青霉 (Paecilomyceslilacinus)除对根结线虫和大豆孢囊线虫的卵具有较强的寄生能力外 ,其代谢产物对多种作物有生理活性 .在番茄幼苗上 30d的试验结果表明 ,以 10 5万孢子 mL浓度的培养滤液 ,促进番茄生长作用最明显 ,地上部鲜重平均比对照高 4g ,根重高0 2g .大豆、小麦、水稻种子对该滤液敏感程度有差异 .大豆最敏感 ,8× 10 7孢子 mL ,80~160倍稀释液就具有促进萌发和生长作用 ,其中 12 0倍稀释液最显著 .其次是水稻种子 ,2 0倍稀释液作用明显 .小麦种子最不敏感 ,10倍稀释液才表现出刺激生长作用 .在各处理中 ,高浓度滤液均相对表现出抑制萌发和生长作用 .培养滤液经柱层析分离收集到 2个蛋白峰 ,SDS-PAGE分析 2种蛋白分子量分别为 12 0 0 0和 2 80 0 0 .用大豆种子 (南农 88- 48)测定这 2种蛋白的生理活性表明均有明显刺激萌发和生长的作用 ,而透析出的培养液 ,同样方法处理大豆种子发现均无调控生长作用 .研究结果表明 ,这种菌的代谢产物的生理活性物质位于蛋白组分中 . The physiological activities of metabolic product of P lilacinus were defermined for variety crops,except for parasitic ability to eggs of root knot nematodes and soybean cyst nematodes.The experimental results shown that the effect of promoting growth of tomato seedling was significant with culture filtrate of the fungus 10 5×10 4spores/mL and fresh weight of plant was 4 g higher than control treafment and fresh weight of the root was 0 2 g. Seeds of soybean,wheat and rice were diffent to the sensitivities of the culture filtrate. Soybean was the sensitivest among of them at dilute 120 times of the culture filtrate and effective at 80 to 160 times of the filtrate.The seed of rice was second and available at dilute 20 times only,the seed of wheat was not sensitive and available at dilute 10 times.Based upon the molecular weights of commercial standards,the two proteins in the filtrate were 12 kD and 28 kD.It was not that the culture solution was effective to growth of soybean,but that the physiological activity elements were in two proteins in the metabolic product of P lilacinus .
