兔出血症-巴氏杆菌病二联灭活苗的研制

Study on Bicombinant Inactivated Vaccine of Rabbit Heamorrhagic Disease and Pasteurellosis

本试验筛选出了具有良好免疫原性的兔瘟强毒株GMH881和兔巴氏杆菌C51-17株,研制出了兔出血症-巴氏杆菌二联灭活苗。确定出二联苗免疫剂量为1.0mL;二联苗在免疫后5d对兔病毒性出血症强毒的保护率达到100%,二联苗免疫后7d对兔巴氏杆菌的保护率均达到80%以上;二联苗免疫家兔后7d兔病毒性出血症HI抗体可达22.75,15d明显上升,二联苗HI效价30d达到最高峰,可达28.0,二联苗在60～120d均可维持在27.0～25.5水平,到180d时抗体水平略有下降,为25.25,二联苗分别在免疫后4个月、6个月用兔出血症强毒攻击,均产生100%保护,在6个月用2个MLD的兔巴氏杆菌攻击,保护率可达70%以上;二联苗4~8℃条件下保存期暂定为1年,25℃条件下暂定为半年。

The trial screened out rabbit hemorrhagic disease virulent strain GMH881 and pasteurellosis strain C51-17 and bicombinant inactivacted vaccine were developed successfully.It was sure that immune dose of the bicombintaint inactivated vaccines were 1.0 mL,the immune protection rate of bicombintaint invactivated vaccine against RHD reached 100% on 5th day vaccination.The immune protection rate of bicombintaint invactivated vaccine against Pm reached over 80% on 7th day after vaccination.After supervising the level of RHDV through HI method,the result showed that the titers of bicombinant inactivated caccine were 22.75,bicombinant propois inactivated vaccine were 25.0 after 7d of injection,and the antibody effect could reach very high level on after the 15th days.which could reach peak level(210.0) on the 30th days.Effect level of bicombinant inactivated vaccine was 27.0-25.5 during 60-120 days,on the 180th day,antibody level reduced 22.25.After four mouth and six mouth post vaccination,the protection rate of the bicombintaint inactivated vaccines was 100% against rabbit haemorrhagic disease with injecting pathogeng,more than 70% against pasteurellosis with 2 MLD of the pasteurlla multocida strain C51-17 on 6th month.The bicombintaint invactivated vaccine can provide effective protection when it was stored at 48℃ for 12 months,at 25℃ for 6 months.
