二次消毒法在大豆无菌苗制备中的应用

摘要 无菌苗的获得是大豆遗传转化中的第一个环节。不同产地的大豆种子可能因其生长环境不同而具有不同程度的带菌率。通过比较不同消毒条件下大豆种子的污染率、发芽率,从而确定一种有效的种子消毒方法。结果表明:不同品种间种子污染率存在显著差异,次氯酸钠液体消毒法对带菌率低的种子消毒有效,经氯气消毒法和二次消毒法处理后的种子污染率明显低于次氯酸钠液体消毒,以二次消毒法对大豆种子的消毒最为彻底;次氯酸钠液体消毒对种子活力影响较大,经氯气消毒和二次消毒的种子活力显著高于次氯酸钠液体消毒。二次消毒法特别适用于带菌率高的种子消毒。 Seed disinfection is the first step in soybean transformation .The seed contamination varied with the environment of their production area .To establish an effective disinfection method ,the contamination ratio and ger-mination ratio under different disinfection treatments were compared in this study .The seed contamination ratio was significant different with their production area .Hypo-chloride sodium liquid disinfection was effective to seeds with low contamination ratio while it affected seed vigor seriously .Chloride gas and double disinfection method can re-move the contaminated pathogen effectively with less affect on seed vigor .Double disinfection can remove the infec-ted pathogen thoroughly and can be applied in disinfection of heave contaminated seeds .
