Effect of Sarsasapogenin on the Biological Hehaviors to Human Gastric Cell Line BGC-823

目的:探讨菝葜皂苷元体外对人胃癌细胞BGC-823增殖、侵袭、粘附的影响及其促进凋亡的作用。方法:采用MTT法、Transwell小室侵袭实验、粘附实验检测菝葜皂苷元对人胃癌细胞BGC-823增殖、粘附和侵袭能力的影响;AnnexinV/PI法检测BGC-823的凋亡。结果:①菝葜皂苷元有抑制人胃癌细胞BGC-823增殖的作用。②菝葜皂苷元作用胃癌细胞BGC-823后,细胞的粘附能力明显降低(P<0.01)。③菝葜皂苷元作用人胃癌细胞BGC-823后,侵袭的细胞数较对照组明显减少(P<0.01)。④菝葜皂苷元能诱导人胃癌细胞BGC-823凋亡。结论:①菝葜皂苷元对人胃癌细胞BGC-823的增殖具有抑制作用。②菝葜皂苷元能抑制人胃癌细胞BGC-823的粘附能力和侵袭能力。③菝葜皂苷元能诱导人胃癌细胞BGC-823凋亡。

This study was designed to explore the effect of sarsasapogenin on the biological behaviors including proliferation,adhesion and invasion capabilities of human gastric cell line BGC-823 in vitro;the effect of sarsasapogenin on the apoptosis of BGC-823;Methods: After human gastric cell line BGC-823 was treated with sarsasapogenin at different concentrations,cell proliferation and adhesion capabilities was assessed using MTT assay.The invasion capabilities of BGC-823 cells were determined using Transwell Method(Boyden Chamber),respectively.Flow cytometry,AnnexinV/PI double staining method was used to observe the early apoptosis induced by different concentrations of sarsasapogenin in human gastric cell line BGC-823.Results: ①Sarsasapogenin significantly inhibited the proliferation of BGC-823 cells.②Sarsasapogenin significantly decreased the adhesion capability of BGC-823 cells(P<0.01).③Sarsasapogenin significantly decreased the invasion capability of BGC-823 cells(P < 0.01).Conclusion: ①Sarsasapogenin inhibited the proliferation of human colon cell line BGC-823.②Sarsasapogenin supressed the adhesion and invasion capability of BGC-823 cells.③Sarsasapogenin can induce the apooptosisof human BGC-823 cells.
