葛根素联合丹皮酚对三种2型糖尿病动物模型胰腺组织变化及葡萄糖转运蛋白4的影响 被引量:2

摘要 目的探讨葛根素联合丹皮酚对3种2型糖尿病动物模型胰腺组织变化及葡萄糖转运蛋白4的影响。方法分别采用特殊膳食诱导法、注射链脲菌素(STZ)、特殊膳食加注射STZ法构建2型糖尿病动物模型,取造模成功的3种Wistar大鼠,每种40只。将各模型随机分为糖尿病(DM)组、单用葛根素(Pue)组、单用丹皮酚(Pae)组和葛根素联合丹皮酚(Pue+Pae)组,每组10只。试验进行8周后测定每种模型的4组大鼠的胰腺线粒体中的ROS、SOD及MDA;Western blot方法检查处理后的肾周围脂肪组织细胞葡萄糖转运蛋白4的含量。结果在实验过程中没有动物死亡,全部进入结果分析。Pue组、Pae组及Pue+Pae组较DM组大鼠胰腺线粒体SOD活性有显著升高(P<0.01),而MDA、ROS水平呈现不同程度降低(P<0.01或P<0.05),其中Pue+Pae组变化更加明显(P<0.01);设定每种模型DM组的脂肪细胞葡萄糖转运蛋白4的吸光度为1,其它组数据与DM组比较,差异均有统计学意义(P<0.05),且Pue+Pae组是DM组的2~3倍(P<0.01)。结论葛根素联合丹皮酚组能降低3种2型糖尿病模型胰腺组织中的MDA、ROS水平、升高SOD水平;葛根素联合丹皮酚治疗组能提高葡萄糖转运蛋白4蛋白的含量。 Objective To investigate the effect of puerarin combined with paeonol on pancreatic tissue and glucose transporter 4 in three models of animal with type 2 diabetes mellitus. Methods 120 Wistar rats modelled by special diet-induced method,injection of streptozotocin( STZ),special dietary plus STZ injection method respectively were randomly divided into 3 parts,40 rats in each part,each part was used to study a model,and each model was randomly divided into diabetic( DM) group,puerarin( Pue) group,paeonol( Pae) group and puerarin combined with paeonol( Pue + Pae)group. After 8-week treatment,ROS,SOD and MDA of mitochondria were measured in 4 rats of each model in the pancreas. Western blot was used to detect the expression of GLUT4 in adipocytes. Results No animal died during the experiment,all were involved in the result analysis. The SOD activity of Pae group pue group and( Pue + Pae) group were all significantly increased( P < 0. 01),while MDA、ROS decreased( P < 0. 01,or P < 0. 05) this change in( pue + pae) group was more obvious( P < 0. 01); the absorbance of GLUT4 of the model group was represented by 1,the other group of data was compared with DM group,there were significant differences( P < 0. 05) and the( Pue + Pae) group was 2 ~ 3 times DM group( P < 0. 01). Conclusion MDA,ROS level were reduced and SOD level was improved; puerarin combined with paeonol treatment group can improve the glucose transporter 4protein content.
