吡咯喹啉醌对兔骨关节炎关节软骨保护机制的研究 被引量:3

摘要 目的研究吡咯喹啉醌(pyrroloquinoline quinone,PQQ)对兔骨关节炎关节软骨的保护机制。方法 6月龄日本大耳白兔36只为实验对象,随机分为实验组、对照组及伪手术组(每组12只)。观察软骨大体形态并行软骨Pelletier JP评分;制备切片行HE染色,光镜下观察HE切片并按照Mankin评分表对其评分;行II型胶原免疫组化染色,光镜下观察II型胶原表达情况;行软骨细胞凋亡检测(TUNEL法),在光镜下观察软骨细胞凋亡情况;通过透射电镜观察软骨细胞线粒体形态和数目的改变。结果实验组和对照组的Kellgren-Lawrance评分差异有显著性(P<0.05);实验组电镜下软骨细胞线粒体数多于对照组。结论关节腔内注射PQQ可保护软骨细胞,从而减轻关节软骨结构及功能的破坏,促进软骨的代谢和自身修复,防止关节软骨退变。吡咯喹啉醌可能是通过保护软骨细胞线粒体而发挥作用的。 Objective To study the protection mechanism of the pyrroloquinoline quinone( PQQ) for osteoarthritis on rabbits. Methods Six months of aged 36 Japanese white rabbits as experimental subjects,were randomly divided into experimental group and control group and pseudo-operation group,each group had 12 rabbits. Knee anteroposterior X-ray film,and in accordance with Kellgren-Lawrance's grading score was observed under an operating microscope articular cartilage of femoral condyle parallel morphological changes in the general rate of cartilage Pelletier JP,HE sections in the light microscope,and in accordance with the Mankin score sheet score, in the light microscope and the apoptosis of cartilage type II collagen immunohistochemical staining and electron microscopy of chondrocytes in the form. Results The experimental group Kellgren-Lawrance score was significantly lower than the control groups( P < 0. 05); The experimental group and the control group cartilage cell mitochondria number had significant difference under electron microscope. Conclusion Intra-articular injection in osteoarthritis of PQQ to protect cartilage cells,thereby reducing the structure and function of articular cartilage damage and promote cartilage metabolism and repair itself. PQQ may be to protect the articular cartilage by protecting cartilage Unitochondria.
