The mechanism of γ-secretase cause Alzheimer’s Disease

摘要 γ-分泌酶剪切C99形成的Aβ在脑中的聚集认为是阿尔茨海默病的元凶,.GXGD结构域",含水腔隙"的发现使得对γ-分泌酶有了进一步认识。本文对γ-分泌酶的结构、作用机制及其抑制剂的研究做综述。 β-amyloid concentration in the brain is considered a major cause of Alzheimer’s Disease. Aβ42,less soluble,is generated via the cleavage of β-amyloid protein precursorbyγ-secretase.New research confirms that a GXGD motif around the activesites of PS,it is important for theγ-secretase- cleavage Because the water-containing-cavityexist,γ-secretase made the intramembrane cleavage in the highlyhydrophobic environment possible.The potential target is theγ-secretase of Alzheimer’s Disease.
