摘要
目的观察复方守宫散对Lewis肺癌小鼠JAK2-STAT3信号通路的影响。方法将55只C57BL/6J小鼠按常规方法接种Lewis肺癌瘤株,6d后选取荷瘤成功的小鼠50只随机分为5组:阳性对照组、西药组、中药高剂量组、中药中剂量组、中药低剂量组,每组10只。阳性对照组:灌服生理盐水0.2mL,1次/d;中药组灌服复方守宫散高剂量组0.6mL(相当于120mg/kg),中剂量组0.4mL(相当于60mg/kg),低剂量组0.2mL,(相当于40g/kg),1次/d;顺铂组按1mg/kg给药,0.1mL/只腹腔注射。每天1次,共14d。在光镜下用HE染色法观察肿瘤组织病理形态学改变情况,用Western Blot法检测JAK2、STAT3蛋白的表达水平。结果各给药组JAK2、STAT3蛋白的表达水平出现不同程度降低,且JAK2降低较为明显。结论复方守宫散能够抑制肿瘤生长,降低Lewis瘤细胞中JAK2、STAT3蛋白的表达,并通过此方式阻断JAK2-STAT3信号转导。
Objective To observe the effects of Compound Shougong Powder on the JACK2-STAT3 signaling pathway of mice with Lewis lung carcinoma. Methods 55 C57BL/6J mice were inoculated with tumor cell lines of Lewis lung carcinoma via a conventional method. 50 tumor-bearing mice were selected after six days and assigned to five groups:a positive control group, a western medicine group, a TCM group with high dosages, a TCM group with medium dosages, and a TCM group with low dosages, with 10 mice in each group. The positive control group was orally given normal saline of 0.2 mL once a day;the TCM groups were orally given compound shougong powder of 0.6mL for the group with high dosages (equal to 120 mg/kg), 0.4mL for the group of medium dosages (equal to 60mg/kg), and 0.2 mL for the group with low dosages (equal to 40 mg/kg) once a day; the Western medicine group was intraperitoneally injected with cisplatin of 1 mg/kg, 0.1mL each, once a day for 14 days. Pathological and morphological changes of tumor tissues were observed via HE staining under light microscope, and expression levels of JAK2 and STAT3 proteins were tested via the Western Blot. Results Expression levels of JAK2 and STAT3 proteins in each group decreased with different degrees, and the decrease of JAK2 was relatively significant. Conclusion Compound Shougong Powder inhibits tumor growth, lowers the expression of JAK2 and STAT3 proteins in Lewis tumor cells, and in such way blocks the JACK2-STAT3 signaling pathway.
