高钼对小尾寒羊肾脏蛋白含量和代谢酶活性及微核率的影响 被引量:6

Changes in kidney protein,metabolic enzyme activities and micronucleus rate in small-tail han sheep induced by dietary high molybdenum

摘要 采用透射电镜、比色法、姬姆萨染色和流式细胞术来探讨高钼对小尾寒羊肾脏蛋白含量、代谢酶活性、微核率及凋亡等的影响。试验采用雄性健康小尾寒羊12只,随机分成3组,分别饲喂含有5.61mg/kg Mo(对照组)、30mg/kg Mo(高钼Ⅰ组)和60mg/kg Mo(高MoⅡ组)的日粮120d(以Na2Mo4·2H2O形式)。随着钼剂量的增加,血清肌酐、血尿素氮、尿酸、肾脏细胞钼含量、微核率以及凋亡率都呈增加的趋势,且显著高于对照组(P<0.01);高钼组小尾寒羊肾脏细胞铜含量、蛋白含量与代谢酶的活性与对照组相比极显著显著降低(P<0.01);透射电镜的结果表明,高钼组肾脏细胞出现不同程度的空泡变性和线粒体肿胀,与肾脏生化指标的观察变化结果相符。结果表明,高钼引起的蛋白含量和代谢酶活性的降低能够影响肾脏细胞的正常功能。 To study the changes in kidney protein,metabolic enzyme activities and micronucleus rate in small-tail han sheep induced by high Mo,transmission electron microscope,colorimetry,Giemsa staining and flow cytometry methods were adopted.12 male small-tail han sheep were divided randomly into 3groups and fed on diet with 5.61mg/kg Mo(Control),30mg/kg Mo(Mo groupⅠ)and 60mg/kg Mo(Mo groupⅡ)in the form of Na2Mo4·2H2O for 120 days,respectively.Compared with the control,concentrations of serum blood urea nitrogen,creatinine and uric acid,and the rate of micronucleus and apoptosis in kidney in small-tail han sheep were increaed significantly(P<0.01),along with the Mo levels.High Mo resulted in lower Cu and protein levels and the metabolic enzymes activites(P<0.01);morphological changes of vacuolar degeneration and swelled mitochondria were ultrastructurally observed in Mo groups,which was consistent with the changes of the indexes of kidney function.These changes in protein and metabolic enzymes caused by Mo could affect the physiological functions of the kidney.
