四神丸对溃疡性结肠炎模型大鼠血清白细胞介素-1β、-4、肿瘤坏死因子-α的影响 被引量:14

摘要 目的观察四神丸对溃疡性结肠炎(UC)大鼠白细胞介素(IL)-1β、-4肿瘤坏死因子(TNF)-α的影响。方法 Wistar大鼠40只分为四组,采用2、4、6-三硝基苯磺酸(TNBS)乙醇灌肠法造模,光镜观察结肠组织病理形态并评分,采用酶联免疫法(ELISA)检测大鼠血清IL-1β、TNF-α、IL-4的水平。结果模型组大鼠结肠组织黏膜层可见溃疡形成,且有明显的炎症、充血存在,证实造模成功。模型组大鼠血清IL-1β、TNF-α含量均高于空白组(P<0.05),而IL-4含量低于空白组(P<0.01);治疗后,四神丸组血清IL-1β、TNF-α水平均较模型组降低(P<0.05、P<0.01),而IL-4含量却显著升高(P<0.01)。结论四神丸能够通过下调IL-1β、TNF-α的表达,升高IL-4的表达,调节肠道异常免疫反应、抑制炎症反应,修复溃疡,进而治疗UC。 Objective To observe the influence of the Si Shen Pill on interleukin-1β( IL-1β),TNF-α and interleukin-4( IL-4) in ulcerative colitis( UC) rats. Methods Experimental rats were divided into four groups. Model rats were made by means of the TNBS / ethanol enema. Colon organization pathological changes were observed by the light microscope. Serum IL-1β,TNF-α and IL-4 were detected by ELISA. Results Ulceration,inflammation,congestion existed in colon mucosa of model rats,which meant the models were made successfully. IL-1β and TNF-α of model rats were higher and IL-4 was lower than those of rats in blank group( P < 0. 05,P < 0. 01). After treatment,compared with the model group,IL-1β and TNF-α were reduced and IL-4 was increased in the Si Shen Pill group( P<0. 05,P<0. 01). Conclusions Si Shen Pill can reduce the IL-1β and TNF-α,increase IL-4,regulate immune abnormalities in intestines,inhibit inflammation,repair ulcers and cure UC.
