摘要
目的:检测micro RNA-126(mi R-126)基因敲减(knock down,KD)小鼠各组织器官中mi R-126的表达,并观察小鼠血糖的变化,初步探讨其意义。方法:分别提取野生型(wild type,WT)和mi R-126 KD模型小鼠中12种组织器官的总RNA,荧光定量PCR探针法检测mi R-126在12种组织器官的表达水平;检测小鼠血糖的变化,并观察小鼠体质量的变化;HE染色观察肝脏和胰腺的病理改变。结果:与正常小鼠相比,mi R-126 KD模型小鼠的脾、胰腺、肝、肌肉和肺等器官组织中mi R-126的表达显著下调(P<0.05);出生第7周和第16周模型小鼠血糖水平均明显增高(P<0.05),HE染色显示模型小鼠胰腺和肝组织呈明显病理异常;出生第13周后,模型小鼠体质量逐渐增加(P<0.05)。结论:mi R-126KD小鼠血糖水平明显增加,可能与胰腺和肝的病理改变有关,提示mi R-126在血糖代谢中具有重要作用,可能与2型糖尿病的发生发展密切相关。
Objective: To detect the expression of mi R-126 in different tissues and organs and the change of peripheral blood glucose in microR NA-126 knock down(mi R-126 KD) mouse, and to explore the pathological signifi cance.Methods: Total RNAs were isolated from twelve kinds of tissues and organs in wild-type mouse(WT) and mi R-126 KD mouse respectively. Then, the expression level of mi R-126 was detected by real-time PCR assay. The levels of peripheral blood glucose and body weight of mi R-126 KD mice were measured. The pathologic changes of pancreas and lung tissue were observed by HE staining.Results: Compared with the WT mice, the relative expression of mi R-126 in spleen, liver, muscle and lung from the miR-126 KD mice were dramatically decreased respectively(P<0.05). The level of peripheral blood glucose in the miR-126 KD mouse increased significantly at seven week and sixteen week after the birth(P<0.05). HE staining showed that the pathological structure of pancreas and liver were abnormal. The body weight of miR-126 KD mice was increased obviously from thirteen week after birth(P<0.05).Conclusion: Peripheral blood glucose levels in the mi R-126 KD mouse were dramatically elevated, which might be related to the pathological changes in the structure of pancreas and liver. These results suggest that mi R-126 may play an important role in the metabolism of blood glucose and the development of type 2 diabetes mellitus.
