摘要
有丝分裂原激活蛋白激激激激酶1(MAP4K1,又名定向造血干细胞激酶1,即HPK1)是一种丝氨酸/苏氨酸激酶。作为MAP4K家族的成员,它与许多接头蛋白,如CARD11,HSI,HIP-55,GRB2家族,LAT,SLP-76家族,CRK家族,BAM32等相联系,参与了各种免疫反应,包括调控细胞增殖与凋亡、抑制TCR/BCR信号和T/B/树突状细胞介导的免疫反应等,进而在自身免疫性疾病、肿瘤和炎症反应中发挥重要作用。
Mitogen-activated protein kinase kinase kinase kinase 1(MAP4K1, also called hematopoietic progenitor kinase 1, HPK1) is a serine/threonine kinase. As a member of the MAP4 K family, MAP4K1 is closely associated with various adaptor proteins such as caspase recruitment domain family member 11(CARD 11), hematopoietic cell-specific protein 1(HS1), HPK1-interacting protein of 55 kD(HIP-55), grow th factor receptor-bound protein 2(GRB2) family, the linker for activated T-cells(LAT), SH2 domain containing leukocyte protein of 76 kD(SLP-76) family, homolog ues of the v-crk oncogene product(CRK) family, the B-cell adaptor molecule of 32 kD(BAM32), etc. It plays impor tant roles in many immunoreactions, such as regulation of cellular proliferation and apoptosis, inhibition of TCR/BCR signaling and T/B/dendritic cells-mediated immune responses. Thus, MAP4K1 is involved in autoimmune diseases and plays a key role in tumor and inflammatory reaction.
