摘要
聚酰胺-胺(polyamidoamine,PAMAM)树状大分子是一类新型的三维结构高分子材料,具有粒径大小可控、单分散性、无免疫原性、低毒性、生物可降解等特点,其作为药物载体具有粒径小、高通透高滞留(enhanced permeability and retention,EPR)效应强、稳定性好、载药量高等优点,通过靶向分子修饰能主动靶向于特定的组织、细胞或某些特定的靶点,达到增效减毒的效果。本文就主动靶向PAMAM给药系统作为药物主动传递平台的优势及其最新研究进展给以综述。
Dendriner polyamide amine(PAMAM)is a new three-dimensional polymer material which possesses properties of controlled particle size,monodispersion,non-immunogenicity,low cytotoxicity and biodegradation.As drug carriers,it has some advantages of small size,high permeability and retention(EPR)effect,good stability,high drug loading and so on,which can actively target specific tissues,cells or some certain targets through modification of targeting molecules,to improve therapeutic effect and decrease toxicity.The advantages and the latest research progress of active targeting PAMAM drug delivery system(ATPDDS)as drug active delivery platform is reviewed.
