新疆额尔齐斯河河鲈肠道寄生的结节三枝钩绦虫的种类鉴定及系统发育关系的研究

Species identification and phylogenic study of Triaenophorus nodulosuslarvae in the perch Perca fluviatilis in Ergis River of Xinjiang,China

摘要 结合形态学和分子生物学方法,对采自新疆额尔齐斯河河鲈肠道的三枝钩绦虫幼虫进行了鉴定。通过对采集的绦虫幼虫的形态特征进行观察,发现虫体头节有2条浅的吸沟,前端有1个不明显的吸盘,盘上有4个三叉形的小钩,初步鉴定为三枝钩绦虫属的种类。通过扩增18SrDNA序列,发现采集的三枝钩绦虫幼虫与GenBank上公布的结节三枝钩绦虫的18SrDNA序列的同源性为99.5%,该序列已在GenBank中提交,获得的登录号为KM272003.1。同时,利用GenBank中三枝钩科的8个属的代表虫种的18SrDNA序列构建系统发育树,发现本研究中采集的绦虫与新西兰和瑞士报道的结节三枝钩绦虫聚在同一分支。上述结果表明,新疆额尔齐斯河的河鲈肠道寄生的三枝钩绦虫幼虫为结节三枝钩绦虫。 Using morphology and molecular biology techniques,the samples of the Triaenophorus larvae parasitizing in the intestines of Perca fluviatilis from Ergis River of Xinjiang,China were identified.The morphological characteristics showed that the scolex has two shallow grooves,with an unobvious sucker,which has four trinacriformed hooklets.So,the larvae were preliminarily identified as Triaenophorus specie.The 18 SrDNA sequence was amplified and the sequence was compared with those of GenBank.In result,the sequence homogeneity was 99.5% with Triaenophorus nodulosus and this sequence was submitted to the database of GenBank.The accession number is KM272003.1.A phylogenetic tree of 8representative species of genera in Triaenophoridae from GenBank was built using 18 SrDNA sequences.We found that the collected tapeworm was in the same branch with T.nodulosus from Oakland(JF514917.1)and Switzerland(Z98404.1).The above-mentioned result showed that the tapeworm found in the intestines of P.fluviatilis in Ergis River was identified as T.nodulosus with morphological characteristics,sequence comparison,and phylogenetic tree analysis.
