玉米赤霉烯酮和脱氧雪腐镰刀菌烯醇及其联合染毒对小鼠肝氧化损伤的研究 被引量:5

摘要 将成年雌性昆明小鼠360只随机分为9组,即空白对照组、脱氧雪腐镰刀菌烯醇(DON)1.5mg/kg(按体重给药,下同)给药组、DON 2.5mg/kg给药组、玉米赤霉烯酮(ZEA)20mg/kg给药组、ZEA30mg/kg给药组、DON 1.5mg/kg+ZEA 20mg/kg给药组、DON 1.5mg/kg+ZEA 30mg/kg给药组、DON 2.5mg/kg+ZEA 20mg/kg给药组、DON 2.5mg/kg+ZEA 30mg/kg给药组,每天腹腔注射1次,每次200μL,连续注射4d。分别在试验的第0、3、5、8、12天测定小鼠肝组织中MDA含量、抑制OH-能力、TAOC、GSH-Px活性和SOD活性。结果,ZEA或/和DON染毒均导致小鼠肝MDA含量明显升高,抑制OH-能力、T-AOC、GSH-Px活性和SOD活性明显降低(P<0.05或P<0.01),具有明显的染毒剂量和染毒时间依赖效应。结果表明,联合染毒具有互作效应或亚加性效应,其中以2.5mg/kg DON与30mg/kg ZEA联合染毒所呈现的亚加性效应最明显。 A total of 360 female healthy Kunming mice were randomly assigned to nine groups.All the mice were continuously injected for 4times in abdominal cavity every 24 hours with different consistency of200μL DON or/and ZEA(Control group,DON 1.5mg/kg,DON 2.5mg/kg,ZEA 20mg/kg,ZEA 30mg/kg,DON 1.5mg/kg+ ZEA 20mg/kg,DON 1.5mg/kg+ ZEA 30mg/kg,DON 2.5mg/kg+ ZEA 20mg/kg,and DON 2.5mg/kg+ ZEA 30mg/kg).We got liver from each group on the 0day(before the first injection),the 3rd day(before the third injection),the 5th day(after the third injection),the 8th day(the fourth day after stop injection),and the 12 th day(the eighth day after stop injection)respectively.Malonaldehyde(MDA),the hydroxy free radical(OH-),total antioxidative capacity(T-AOC),activity of glutathione peroxidase(GSH-Px)and activity of superoxide dismutase(SOD)were analyzed.In result,DON and/or ZEA markedly increased the levels of MDA,significantly decreased the inhibition capacity of OH-,TAOC,the activity of GSH-Px and SOD(P<0.05 or 0.01),and the effects were time and dose dependent.The result showed that the mixture exposure could exacerbate the toxicity or the additive effect,and the additive effect was the most obvious with 30mg/kg ZEA and 2.5mg/kg DON.
