Keap1 affects cell motility by regulating filament actin reorganization

摘要 目的:探讨抑癌基因Keap1在细胞运动中的作用及其调控机制.方法:构建真核表达载体pIRES-EGFP-Keap1,应用Lipofectamine 2000脂质体转染COS-7和A549细胞并以其作为实验组,以其空载pIRES-EGFP转染细胞组为对照组,未转染组为正常组.免疫印迹检测两组细胞Keap1蛋白表达水平;采用划痕实验观察细胞迁移能力,免疫荧光显色显示细胞骨架F-actin的解聚与重组.结果:已知COS-7细胞不表达Keap1,转染pIRES-EGFP-Keap1后,免疫印迹显示Keap1蛋白表达明显增加,提示载体构建和转染成功.细胞迁移实验显示Keap1过表达使COS-7细胞迁移能力明显下降,过表达Keap1的COS-7和A549细胞内F-actin纤维束增多、增粗、增长,交错排列横穿整个胞质;对照组束状F-actin纤维较少且短,在细胞质散在分布.结论:Keap1过表达能够促进细胞内应力纤维形成,增加细胞骨架稳定性,发挥抑制细胞运动的作用.
