脐血干细胞移植联合电针治疗促进脊髓损伤神经功能恢复 被引量:1

摘要 目的:观察脐血干细胞移植联合督脉电针治疗对脊髓全横断损伤大鼠神经功能的恢复作用.方法:建立SD大鼠胸段脊髓完全横断模型,随机分为损伤模型组、脐血干细胞移植组(脐血组)、督脉电针治疗组(电针组)和脐血干细胞移植联合督脉电针治疗组(联合组);梯度离心法分离人脐血单个核细胞,尾静脉注射移植;取大椎、命门二穴,电针治疗.应用Basso-Beatt ie-Bresnahan(BBB)评分评价各组大鼠后肢功能的恢复情况;各组分别于7、14和28d3个时间点检测大脑皮层运动诱发电位;电镜观察脊髓髓鞘修复及再生情况.结果:各组脊髓神经功能均有不同程度恢复.联合组动物后肢功能恢复显著,BBB评分与其他3组比较差异存在;联合组皮层运动诱发电位的潜伏期缩短、波幅增高,与其他3组比较差异存在.电镜观察,联合组髓鞘松散程度明显减轻,结构规整,线粒体肿胀减轻,薄髓的再生纤维明显增多.结论:脐血干细胞移植联合督脉电针治疗对脊髓损伤的功能恢复有促进作用. Objective:To investigate the therapeutic effect of transplanted human umbilical cord blood stem cells(hUCBSC) in combination with electro-acupuncture on neural functional recovery of injuried spinal cord in rats.Methods:Adult SD rats were subjected to the transected thoracic spinal cord injury(SCI) model,and divided into four groups randomly:control group,stem cell group,electro-acupuncture group,stem cell combined with electro-acupuncture group(combination group).The hUCBSC cells were isolated from umbilical cord blood by gradient centrifugation,and injected intravenously through the tail vein.Electro-acupuncture was performed in Dazhui(DU14) and Mingmen(DU4) of DU channel.Recovery of hind limb locomotor function was assessed based on Basso-Beattie-Bresnahan(BBB) scores.The motor evoked potentials(MEP) in the cortex were detected by electrophysiological method at 7,14,28 days.Regeneration and repair of spinal cord were observed under the electron microscope.Results:spinal cord function in each group recovered at different degrees.The BBB score of combination group was significantly different from other three groups,and the movement of the posterior limb recovered significantly.The latent period was shorter and amplitude of MEP was higher in the combination group significantly than that in other three groups.The myelin sheath was arranged more regularly,the degree of swelling mitochondria was reduced and the thin regenerated myelin sheaths increased in the combination group significantly.Conclusion:hUCBSC transplantation combined with electro-acupuncture contributes to the functional recovery of injuried spinal cord.
