摘要
目的:检测基质细胞衍生因子CXCL 12与Wnt信号通路中阻断剂DKK1在多发性骨髓瘤的表达和相互作用.方法:收集30例诊断为多发性骨髓瘤患者和18例健康对照组血清;采用ELISA方法测定血清CXCL12、DKK1水平;体外培养多发性骨髓瘤细胞株,分别加入30 ng/ml CXCL12和100 ng/ml DKK1,免疫印迹观察DKK1的表达量.结果:多发性骨髓瘤患者血清CXCL12水平显著高于健康对照组,DKK1水平亦显著高于健康对照组.且多发性骨髓瘤患者CXCL12和DKK1水平呈正相关,健康对照组无这种相关性.免疫印迹结果显示,体外培养细胞株在CXCL12介导下,DKK1的表达量较对照组及CXCL12组都有明显升高.结论:CXCL12与DKK1呈正相关性,共同介导多发性骨髓瘤.
Objective:To detect the expression and interaction of DKK1 in the multiple myeloma bone as a blocker in the CXCL12 and Wnt signal pathway.Methods:Clinical data and serum were collected from 30 cases diagnosed as multiple myeloma and 18 cases as healthy control.ELISA method was used to examine the level of CXCL12,DKK1 in serums 30 ng/ml CXCL12 and 100 ng/ml DKK1 were added separately into the multiple myeloma cell lines cultured in vitro.Western blotting was used to observe expression of DKK1.Result:CXCL12 level in serum of people diagnosed with multiple myeloma was significantly higher than healthy control(3 468.5 ± 1 330.8) pg/ml vs(2 987.6 ±546.9) pg/ml.DKK1 in serum of multiple myeloma had also a significantly higher level than that of control(3 657.8±3 330.9) pg/ml vs 1 870.4±646.7) pg/ml).CXCL12 and DKK1 level in multiple myeloma had a positively correlated relationship,while there was no positively correlated relationship between CXCL12 and DKK1 level in the healthy group.Western blotting showed that mediated by CXCL12,the expression of DKK1 increased significantly in camparison with that in the control and CXCL12 group.Conclusion:CXCL12 has a positive relationship with DKK1,mediating multiple myeloma together.
