血清-糖皮质激素诱导激酶1基因对少突胶质细胞分化的影响

摘要 目的:探讨血清-糖皮质激素诱导激酶1(SGK1)对少突胶质细胞分化的影响.方法:利用免疫荧光染色检测糖皮质激素对体外培养少突胶质前体细胞(OPCs)分化的影响;原位杂交检测SGK1在少突胶质细胞中的表达情况;通过构建SGK1真核表达载体和鸡胚神经管电穿孔转化,研究SGK1过表达对少突胶质细胞发生发展的影响.结果:在体外,SGK1的激活物氢化可的松能促进OPCs分化;抑制物GSK650394则会阻断OPCs分化,鸡胚神经管过表达SGK1会促进少突胶质细胞转录因子2和Sox10的提前表达.结论:SGK1能促进少突胶质前体细胞的发生及分化. Objective:This study aimed to explore the differentiation of oligodendrocytes regulated by serum and glucocorticoid inducible kinase 1(SGK1).Methods:Immunofluorescence staining was used to analyze the role of glucocorticoids on differentiation of oligodendrocyte precursor cells(OPCs) in vitro.In situ hybridization was used to detect the expression of SGK1 in oligdendrocytes.SGK1 overexpression vector construction and the electroporation were used to detect its overexpression which led to impact on oligodendrocyte development in the chick neural tube.Results:SGK1 activator hydrocortisone could promote the differentiation of OPCs,but the SGK1 inhibitor GSK650394 could block its differentiation in vitro.Overexpression of SGK1 promoted the early expression of Olig2 and Sox10 in the chick neural tube.Conclusion:SGK1may promote the generation and differentiation of oligodendrocyte precursor cells.
