摘要
以大丽花嫩芽为外植体进行了组织培养的研究。结果表明:在培养基MS+0.5mg/L6-BA+0.1mg/L NAA诱导顶芽生长和腋芽萌发最适宜。萌发的不定芽在增殖培养基上诱导增殖,在培养基MS+0.5mg/L6-BA+0.1mg/L NAA上增殖效果最好,增殖系数达6.4,且不定芽生长健壮。茎段比叶片更容易诱导出愈伤组织,最高诱导率为50%,所用培养基为MS+0.2mg/L6-BA+0.5mg/L2,4-D。最适宜的生根培养基为不加任何激素的MS培养基。
Using tender sprouts as the explants, the tissue culture of Dahlia pinnata. was studied. The results showed the optimum medium for shoot tip and axillary-buds sprouting were MS and 0.5mg/L 6-BA and 0.1mg/L NAA. The media of MS and 1.0mg/L KT and 0.1mg/L NAA were successful for proliferation of plantlets, and each plant produced up to 4.5plantlets. the stalk induced callus more easily than blade, the optimum medium were MS and 0.2mg/L 6-BA and 0.5mg/L 2,4-D, and the sprouted ratio reached to 50%. The medium of MS without any hormone is good for rooting.
