抗β2-mG杂交瘤细胞系的制备及其单克隆抗体应用研究

摘要 本文应用纯化的β2-mG免疫BALB/c小鼠,制备了11株抗β2-mG单克隆抗体(McAb)的杂交瘤细胞系。首次应用β2-mG McAb建立了竞争性、宽范围(0.31~10μg/ml)的β2-mG放射免疫测定法(RIA),并将此法与市售商品药盒多克隆抗体(PcAb)RIA(检测范围0.005~0.08μg/ml)进行了比较,两者之间有极显著正相关(P<0.001)。实验结果表明:β2-mG McAb完全可以替代PcAb。作者又采用β2-mG McAb,将生物素-亲合素系统(BAS)引入时间分辨免疫荧光测量法(TRIFMA),成功地在国内首次建立人血清β2-mG的BAS-TR-IFMA,其可测范围达1.25-40μg/ml。 In the present study, 11 different McAbs were obtained by immunization of BALB/c mice with purified β2-mG antigen. A competitive radioimmunoassay (RIA), which was of a wide detecting range (0.31-10μg/ml),was established using β2-mG McAb and compared with RIA using polyclonal antibodies (detecting range of 0.005-0. 08μg/ ml). The findings showed that inter-assay correlation coefficient was 0.979 (p<0.01). Our results demonstrated that β2-mG McAb may fully replace polyclonal antibodies in RIA. Moreover, human serum β2-mG BAS-TRIFMA was established using β2-mG McAb and BAS, which had a detection range of 1.25-40 μg/ml.
