摘要
本文以单核细胞衍生的基因重组人IL-8(MIL-8)为免疫原,经常规免疫、融合、克隆化制备了6株抗MIL-8的单克隆抗体(2G2、4E1、4C3、4D7、5C9、5A4)。其中5株(除4D7)对MIL-8的嗜中性粒细胞趋化活性具有中和作用,6株单抗均与内皮细胞衍生的重组人IL-8(EIL-8)具有交叉反应,但与表达IL-8的载体菌裂解液均无交叉反应。
Six monoclonal antibodies designated 2G2, 4C3, 4E1, 4D7, 5A4 and 5C9 were obtained by fusion of Sp2/0 myeloma cells with splenocytes from mouse immunized with rHuIL-8. Among them, 5 McAbs can neutralize the chemotactic activity of IL-8 to neutrophils. All of them can cross-react with EIL-8. However, none can cross-react with lysate of carrier bacteria used to express IL-8.
