摘要
本文以人胃癌SGC-7901细胞为实验模型,应用胶体金示踪技术,对鼠抗人胃癌单克隆抗体KGb_2的吸附内化与液相胞饮之间的关素进行了探讨。电镜观察发现:(1)液相胞饮的入胞途径有被覆小凹、微胞饮凹陷及大胞饮凹陷3种。抗体的吸附内化入胞途径与之一致;(2)凡是抑制液相胞饮活动的因素均能抑制MGb_2抗体的吸附内化。根据本研究结果及对相关文献材料的分析,本文提出抗肿瘤抗体的吸附内化依赖于膜胞饮功能区这一内化细胞生物学理论的新观点。
Utilizing colloidal gold tracing technique, we investigated the relationship between adsorptive internalization of an antigastric cancer monoclonal antibody MGb2 and fluid phase pinocytosis in the model cell line of human gastric cancer SGC-7901, Electron microscopy indicated that: (1) the entry routes of fluid phase pinocytosis were coated pits, micro-and macropinocytic invagination, which appeared identical to the entry routes of adsorptive internalization of MGb2 antibody; (2) factors inhibiting fluid phase pinocytosis also retarded adsorptive internalization of MGb2 antibody.These findings suggest that adsorptive internalization of antitumour antibodies may be dependent on the membrane pinocytosis domains.
