摘要
本文利用氯胺T碘标技术对肺癌单抗(McAb 3D3)体外结合特性进行分析,McAb 3D3与肺腺癌细胞株L342的结合常数为3.2×10~8mol^(-1),每个L342细胞表面McAb 3D3的平均结合位点数为2.5×10~6个。RIA观察各种处理因素对L342细胞表面结合位点与^(131)I-McAb 3D3结合抑制率的影响,1%NaIO4、0.2%蛋白酶K、100%甲醇和100%丙酮的结合抑制率分别为72%、46%、14%和16%。结果提示,McAb 3D3所针对的3D3抗原是一种糖蛋白。
The in vitro, binding characteristics of monoclonal antibody (McAb) against lung adenocarcinoma L342 cells, McAb 3D3, were studied by the chloramine-T (Ch-T) Ⅰ-radiolabelling method. The association constant of M,cAb 3D3 is 3.2×108 mol and the number of its binding sites per L342 cell is 2.5×106. The inhibiting effects of various drugs on the binding ability of these sites on L342 cells were observed by radioim-munoassay (RIA). The findings revealed that the binding inhibition rate of 1% NaIO4, 0.2% proteinase K, 100% methanol and 100% acetone was 72%, 46%, 14% and 16%, respectively, suggesting that the antigen recognized by the McAb is a glycoprotein.
