摘要
用转AtHSP1 8 2 promoter/GUS基因的转基因拟南芥为材料 ,体内 (invivo)实验证明钙 钙调素调节热激基因的表达。结果表明 ,无论在热激还是非热激条件下 ,CaCl2 处理都能提高转基因拟南芥的β 葡萄糖苷酸酶 (GUS)的活性 ,而钙离子螯合剂EGTA、钙离子通道阻断剂LaCl3和异博定、钙调素拮抗剂W 7、氯丙嗪 (CPZ)和三氟拉嗪 (TFP)的处理均使 β 葡萄糖苷酸酶的活性下降。
The involvement of calcium and calcium-activated calmodulin (Ca 2+-CaM) in heat shock signal transduction in Arabidopsis (Arabidopsis thaliana) was investigated. Using the transgenic Arabidopsis plants which have the AtHsp18.2 promoter/GUS gene fusion,it was found that the level of β-glucuronidase (GUS) activity was up-regulated by the addition of CaCl 2 and down-regulated by the calcium ion chelator EGTA,the calcium ion channel blockers LaCl 3 and verapamil,or the CaM antagonists N-(6-aminohexyl)-5-chloro-1-naphthalenesulfonamide (W7),chlorpromazine (CPZ) and trifluopetazine (TFP). CaCl 2 not only increased the expression of GUS activity after HS,but also up-regulated the expression of GUS under non-HS conditions.These results provide further in vivo proofs of the involvement of the Ca 2+-CaM signaling system in HSP gene expression.
