摘要
本研究应用免疫细胞化学PAP漂浮法和计算机图像分析技术,半定量研究了烫伤对大鼠下丘脑弓状核β-内啡肽神经元分泌功能的影响。结果表明,烫伤后即刻、1 h弓状核β-内啡肽神经元数目和阳性反应面积均明显减少(P<0.05),而弓状核下行到正中降起部的阳性纤维串珠状结构却略有增加。提示大鼠烫伤后即刻和1 h从下丘脑弓状核经垂体门脉系进入血液的β-内啡肽量增加。至烫伤后2、3、4h,弓状核β-内啡肽神经元数目和阳性反应面积与对照组相比无明显差别,说明其细胞分泌和合成功能趋向平衡。
Effect of scalding on secretory function of beta-endorphin neuron in the arcuate nucleus of the rat hypothalamus was studied semiquantitatively using immunocytochemistry and image analysis. The results showed that the number of beta-endorphin neurons and area of positive products reduced significantly (P<0.05) at 0h and 1 h after scalding, while the number of positive varicose-like fiber in the median eminence descending from the arcuate nucleus increased slightly. These results implied that the amount of beta-endorphin in the blood, which came from the arcuate nucleus of the rat hypothalamus through hypophyseal portal system, increased at 0 h and 1 h after scalding. And at 2, 3, 4h after scalding the number of beta-endorphin neurons and area of positive products in the arcuate nucleus were not different significantly compared with those of the control group, indicating that the rates of secreting and synthesizing of beta-endorphin in the neurons were paralleled.
