转化条件对质粒DNA转化大肠杆菌的影响 被引量:10

The Influence of the Transformation Conditions on the Transformation Frequency of E.coli by Plasmid DNA

摘要 研究了质粒DNA大小、质粒DNA浓度、CaCl2 浓度、热休克时间及感受态细胞保藏时间等因素对大肠杆菌HB1 0 1和JM1 0 5转化频率的影响 ,并对转化子中质粒DNA进行了分离、酶切、琼脂糖凝胶电泳检测。结果表明 ,CaCl2 浓度、质粒大小和浓度 ,以及感受态细胞的活力对转化频率有重要影响 ,42℃热休克处理可以提高转化频率。 The factors effecting on the transformation frequency of E.coli HB101 and JM105 by plasmid DNA were studied.The results show that the concentration of CaCl 2,the size and the amount of plasmid DNA,and the vitality of the competent cell have significantly effects on the transformation frequency respectively.The transfomation frequency can be increased by heat shock in 42℃.The plasmid DNA in the transformant has been isolated and analysised with the electrophoresis of agarose gel.
