摘要
目的 :观察高压氧 (HBO)对异基因骨髓移植小鼠血清IL 2 ,TNF α ,IL 4及IL 10浓度的影响 ,探讨HBO防治急性移植物抗宿主病 (aGVHD)的机制。方法 :受鼠移植前接受高强度预处理。HBO组移植前 3d及移植后 2周接受高压氧处理。 2周后用ELISA法检测受鼠血清细胞因子的浓度。结果 :对照组均出现aGVHD ,而HBO组没有出现aGVHD。HBO组血清IL 2和TNF α浓度均明显低于对照组 (P <0 .0 5 ) ;而IL 4和IL 10则明显高于对照组 (P <0 .0 5 )。结论 :HBO可有效预防异基因骨髓移植小鼠出现aGVHD ,其机制可能与HBO显著降低受鼠血清IL 2和TNF α浓度及升高IL 4和IL 10浓度有关。
Objective: To observe the effect of hyperbaric oxygen(HBO) on levels of IL-2,TNF-α,IL-4,and IL-10 in allogenic bone marrow transplantation(BMT) and to study the mechanism of HBO antagonizing acute graft-versus-host disease(aGVHD).Methods:The recipient mice were exposed to total body radiation of 1 800 cGy for 10 hours.The HBO group was treated with HBO 3 days before BMT and 2 weeks after BMT.Levels of these cytokines were determined by using ELISA method.Results:All mice in the control group suffered from aGVHD,but none in HBO group.Compared with the control group,levels of IL-2 and TNF-α in HBO group decreased significantly( P< 0.05),and levels of IL-4 and IL-10 increased significantly( P< 0.05).Conclusion:Hyperbaric oxygen can prevent aGVHD.And the mechanism is relevant to the effect of HBO on the cytokines.
