硬脂酸、苹婆酸对培养的大鼠肝癌细胞膜表面Mg^(2+)-ATP酶活性的影响 被引量:1

摘要 作者应用细胞化学方法研究了培养的大鼠肝癌FSK-7902细胞,在硬脂酸、苹婆酸处理后,细胞膜表面Mg^(2+)-ATP酶的活性变化.并用显微分光光度计进行了定量分析.结果显示,未处理的培养大鼠肝癌细胞膜表面Mg^(2+)-ATP酶的阳性反应十分显著,主要分布于细胞与细胞相互接触面上.硬脂酸、苹婆酸处理后的癌细胞,当呈现生长抑制状态时,细胞相互接触面上Mg^(2+)-ATP酶的阳性反应显著降低.尤其在硬脂酸和苹婆酸联合应用时,酶阳性反应降低更显著.分光光度计测定结果表明,硬脂酸、苹婆酸处理后,癌细胞膜表面酶的总光密度和平均光密度都明显降低.提示细胞膜Mg^(2+)-ATP酶的活性降低可能与细胞产生生长抑制有关.硬脂酸和苹婆酸联合应用有协同作用,对其机理进行了讨论. Mg2+ - ATPase activity of the rat hepatoma FSK-7902 cells in culture was investigated by cytochemical method. The cytochemical reactions of the rat hepatoma cell membrane were quantitated by microspectrophotome-ter. The results showed that the positive reactions of the enzyme were located intensively on the contact surlaces of the untreated hepatoma cells in confluent monolayer culture. The hepatoma cells treated with stearic acid, sterculic acid or their combination showed the decrease of Mg2+ -ATPase positive reactions, in particular, on the contact surfaces of the adjacent cells. The Mg2+-ATPase positive reactions on the hepatoma cell membrane was less intensive after treatment with combination of stearic acid and sterculic acid than after treatment with either acid alone. The total optical density value and mean optical density value were decreased obviously after treatment with stearic acid, sterculic acid or their combination.
