摘要
应用免疫荧光细胞化学方法观察了体外LAK细胞杀伤人直肠腺癌细胞(HR8348)时效靶细胞内微管蛋白的变化。结果表明,LAK细胞与靶细胞相互接触形成效靶复合体后,两种细胞胞浆内微管蛋白出现重排现象。LAK细胞内的微管蛋白全部集中在两类细胞质膜接触处,呈月芽状分布。靶细胞内的微管蛋白在效靶复合体质膜接触处浓集成斑块状并与LAK细胞的月芽状结构融合,靶细胞逐渐变性坏死。这提示,靶细胞的溶解可能与效靶细胞的微管重排有关。
The human rectal adenocarcinoma cell line ( HR8348 ) was treated with lymphokine activated killer ( LAK ) cells in vitro and the changes of the microtubulin in both the effector and target cells were investigated, with the aid of immunofluorescence microscopy.It was revealed that after the attachment of LAK and tumor cells, effector-target conjugates were formed. and the mocrotubulin, in both the effetor and the target cells redistributed .In the LAK cells, the microtubulin concentrated mainly in the contact region,forming a crescent -like structure, while in the target cells, the microtubulin condensed into patches and fused with the crescent-like structure of the LAK cells.Eventually, the target cells degenerated and died. It was suggested that the lysis of the target cells may be related to the rearrangement of the microtubulin in both the effector and target cells.
