摘要
从日本冲绳县土壤采样经垃圾处理场堆肥化处理,分离到一株有固氮能力的菌株HS-G8,该菌为革兰氏阳性菌,不产芽孢,无运动性.在LB固体培养基上,菌落为黄色,圆形,边缘整齐,有光泽.有明显的杆-球状的生长周期,(1d内呈杆状,2d以上呈球状).对该菌株HS-G8的16SrDNA的全序列测序并进行DNA同源性分析,发现它与Arthrobacter属的关系最近,与Arthrobacternicotianae的同源性为99.1%.其主要生理生化特征也与Arthrobacter属相符.初步鉴定HS-G8可能为Arthrobacter属的一个新种.
A bacterial strain HS-G_8 was isolated from the garbage heap in Japan, gram-positive, non-endospore, non-motile, azotobacter. The colonies on LB plate tended to appear yellow-pigment glossy circulars. Obvious growth period of bacilliform-micrococcus was observed. The full-length of 16S rDNA sequence of strain HS-G_8 was determined and analyzed. Strain HS-G_8 was most related to the genus of Arthrobacter. The homologous of nucleartides between HS-G_8 and Arthrobacter nicotianae was 99.1% and the main biochemical and physiological features of strain HS-G_8 were very similar to that of Arthrobacter. Since nitrogen-fixtion has not been observed previously among Arthrobacter sp., strain HS-G_8 was identified as a new Arthrobacter species which hold the ability of nitrogen-fixtion.
