摘要
以长春花(Catharanthusroseus(L.)G.Don)幼嫩叶片作为外植体,接种于附加不同浓度的NAA,2,4-D、6-BA、KT及其组合的MS固体培养基上,结果发现除单独使用KT不能产生愈伤组织外,其它3种单独使用时,在一定的浓度范围均有愈伤组织产生;最佳的组合培养基为MS+NAA0.5mg/L+2,4-D0.5mg/L+6-BA2.0mg/L.进一步研究发现,在黑暗和光照培养条件下,愈伤组织增殖呈"S"型,且生长周期均为27d.
Young leaves of Catharanthus roseus (L.) G. Don which were used as explants, were cultured in medium MS supplement with different kinds, different concentrations and combinations of plant growth regulators. The results showed that: there was no callus when medium MS supplied with only KT. MS+NAA 0.5 mg/L+2,4-D(0.5 mg/L)+6-BA 2.0 mg/L was the optimum medium for callus induction. When cultured in the dark and the white light, the callus growth circles were about 27 days, and the growth curves were in the shape of 'S'. The cells grew more rapidly in the white light than in the dark.
