高血压发生、发展过程中动脉神经肽Y-Y1受体数量变化的实验研究 被引量:2

Experimental study on the quantity of expression of Neuropeptide Y-Y1 receptors on the arterial wall during development and progression of hypertension

摘要 目的 探讨动脉壁上的神经肽Y(NPY) Y1受体在高血压发生、发展过程中的变化。方法 实验用SHR和WKY大鼠 (每组各 5只 )。采用免疫荧光法检测分离血管的NPY Y1受体 ,应用荧光显微镜观察、取像。结果 从形态学的角度证实NPY Y1受体存在 ,并发现其在小动脉上的分布特点为间隔条带状分布。在脑部动脉中 ,随着血管直径的减小 ,NPY Y1受体表达量增加。在发生高血压前 ,SHR大鼠的NPY Y1受体已高于WKY大鼠 ,出现高血压后 ,其表达量反而下降 ,但仍高于WKY大鼠。结论 高血压的靶血管损害主要在阻力血管 ,而NPY Y1受体主要分布于小血管 ,NPY Y1受体的这种分布特点可能是高血压形成并损害血管的一种机制。 Objective To observe the changes of Neuropeptide Y-Y1(NPY-Y1) receptors distributed on the arterial wall during development and progression of hypertension. Methods The cerebral arteries were isolated from SHR and WKY rats, respectively, and then the immunofluorescence technique was performed to detect the changes in expression of NPY-Y1 receptors on the arterial wall. Results The experiment confirmed the existence of the NPY-Y1 receptors on the wall of the arteries. The distribution of NPY-Y1 receptors was characterized by presence of band-like septa especially on the wall of the small arteries. The smaller the diameter of the artery, the more abundant the NPY-Y1 receptors appeared. The expression quantity of NPY-Y1 receptors in the SHR increased before the occurrence of hypertension, but decreased after its emergence, yet still more than that in the age-matched WKY groups. Conclusions The target blood vessel impaired by hypertension is mainly the resistance vessel. NPY-Y1 receptors are mainly distributed on the small arterial wall, hence this indicates that more expression of NPY-Y1 receptors may be one of the causes of hypertension.
