Expression of ACTH receptor in human lymphoid tissues

摘要 目的 通过检测ACTH受体及其mRNA在人淋巴组织中的表达情况 ,寻求神经内分泌系统 免疫系统之间功能双向调节的形态学依据。方法 应用免疫组化SP法及核酸分子原位杂交法检测ACTH受体及其mRNA在人淋巴结、脾脏及肠道淋巴组织中的表达。结果 免疫组化显示ACTH受体在人各种淋巴组织中表达的阳性率为 82 .5 % ,与原位杂交阳性率 70 .0 %之间的差异无显著性。结论 人的免疫活性细胞不但可以表达ACTH受体蛋白 ,还可以合成其mRNA ,ACTH受体为神经内分泌系统 免疫系统网络共有的生物学语言媒介 。 Objective To find the morphological evidences of the bi-directional regulation between neuroendocrine system and immune system by detecting the expression of ACTH receptor protein and its mRNA in human lymphoid tissues. Methods ACTH receptor and its mRNA were assayed in human lymph node, spleen and intestinal lymph tissuebyimmunohistochemicalSPmethodand in situ hybridization respectively. Results Immunohistological staining showed that 82.5% of lymphoid tissues yielded positive expression of ACTH receptor; in situ hybridization showed that 70.0% of lymphoid tissues yielded positive expression ACTH receptor mRNA; there was no significant difference between the positive rate of in situ hybridization and the positive rate of immunohistochemical staining. Conclusion Human immunoactive cells not only express the ACTH receptor protein, but also synthesize its mRNA. ACTH receptor is one of the common biologic mediators between neuroendocrine system and immune system, both endocrine-derived ACTH and immune-derived ACTH may regulate the function of immune system by ACTH receptor on the membrane of immunoactive cells.
