摘要
背景:蛙皮素具有刺激多种正常和肿瘤细胞增殖的作用。目的:探讨蛙皮素在化学致癌剂N鄄甲基鄄N'鄄硝基鄄N鄄亚硝基胍(MNNG)诱发人胃黏膜上皮永生细胞系GES鄄1癌变过程中的作用。方法:经蛙皮素和(或)MNNG处理后,观察各组GES鄄1细胞的光镜和电镜下形态学改变,采用四甲基偶氮唑蓝(MTT)比色法检测细胞增殖速度,软琼脂集落形成实验检测转化细胞的特性,流式细胞仪检测细胞周期蛋白(cyclin)D1的表达。结果:GES鄄1细胞经蛙皮素与MNNG共同处理后传至20代以上时,细胞增殖速度较MNNG单独处理的细胞加快,cyclinD1表达增高,而软琼脂集落形成率与MNNG单独处理的细胞相比无显著差异;蛙皮素与MNNG共同处理的GES鄄1细胞形态学表现为细胞排列紧密,细胞核增大,核仁增多,细胞质减少,较MNNG单独处理的细胞改变明显。结论:蛙皮素可与化学致癌剂协同促进正常上皮细胞癌变,其机制可能与cyclinD1的过表达有关。
Background: Bombesin can accelerate the proliferation of many kinds of normal and cancer cells. Aims: To investigate the effects of bombesin on the malignant transformation of immortalized human gastric epithelial cell line GES-1 induced by chemical carcinogen N-methyl-N′-nitro-N-nitrosoguanidine (MNNG). Methods: GES-1 cells were treated by bombesin or MNNG only, or both of them. In each group, the morphologic changes of GES-1 cells were observed under light and electron microscope; the cell proliferation was evaluated by methyl thiazolyl tetrazolium (MTT) colorimetric assay; the characteristics of transformed cells were examined by colony formation in soft agar; the cyclin D1 expression was detected by flow cytometry. Results: After treatment with bombesin and MNNG, and the passage number was over 20, the proliferation and cyclin D1 expression of GES-1 cells were increased, as compared with the cells treated with MNNG only. The colony formation in soft agar was not significantly different between the two groups, but their morphologic changes were obviously different. The GES-1 cells treated with bombesin and MNNG became closely arranged, the nuclei enlarged, the number of nucleoli increased, and the cytoplasm reduced in volume. Conclusions: Bombesin seems to exert a synergistic effect with chemical carcinogen in promoting malignant transformation of normal epithelial cells, which may be related to the increased cyclin D1 expression.
