摘要
采用TBA检测方法 ,体外观察了CdCl2 对大鼠线粒体脂质过氧化产物丙二醛 (MDA)生成的毒性作用和对半胱氨酸 -Fe2 + 诱导的线粒体脂质过氧化的影响。结果表明 ,1 μmol/L~1mol/LCdCl2 均能诱导肝线粒体的脂质过氧化 ,使MDA含量增加 ,但只有CdCl2 浓度高于 1mmol/L时 ,诱导作用才与对照组有显著性差异。在Fe2 + Cd2 + 半胱氨酸共同对肝线粒体作用时 ,由于试剂加入顺序的不同 ,1 0 0~ 1 0 0 0mmol/LCdCl2 对线粒体脂质过氧化可产生诱导或拮抗作用。结果提示 ,CdCl2 可直接诱导线粒体脂质过氧化 ,在做几种金属离子体外协同对肝脂质过氧化影响的实验时 ,应考虑试剂加入顺序所产生的不同影响。
The effects of CdCl 2 on rat mitochondria and the lipid peroxidation in mitochondria induced by Fe 2+ -cysteine were observed in vitro by using test method of TBA.The results show that 1 μmol/L~1 mol/L CdCl 2 can induce lipid peroxidation of rat mitochondria in vitro,increased MDA.But only when the density of cadmium chloride is over 1 mmol/L,the induction is significant different compared with the control.Because of different orders of test samples,when Fe 2+ -Cd 2+ -cysteine act together on liver mitochondria,100~1*!000 mmol/L CdCl 2 may bring about induction or antagonistic action to the lipid peroxidation in mitochondria.The results also show that since cadmium chloride directly induces lipid proxidation in mitochondria,the effects of different orders of test samples must be taken into account while making experiments with metal ions that work together in vitro on lipid peroxidation.
