山羊骨髓间充质干细胞的分离培养及向神经细胞诱导分化的研究 被引量:5

摘要 采用体外细胞培养技术将山羊胎儿骨髓中的干细胞从骨髓血中分离并培养扩增;分别用含β 巯基乙醇或当归的培养液诱导骨髓间充质干细胞分化为神经细胞,免疫组织化学鉴定神经丝蛋白和神经元特异性烯醇化酶的表达。结果表明,通过贴壁培养法,可使低丰度的山羊骨髓间充质干细胞在体外实现分离扩增;诱导后的细胞强表达神经丝蛋白和神经元特异性烯醇化酶。还建立了一种简单易行的体外分离与培养山羊骨髓间充质干细胞的方法,同时表明山羊骨髓间充质干细胞同样具有分化为神经细胞的潜能,为今后对某些疾病的治疗提供了理想的动物模型。 To culture bone mesenchymal stem cells(BMSCs) in vitro ,and to induce them into neurocytes,BMSCs of goat were extracted from bone marrow and purified based on density centrifuge,then expanded in culture medium. BMSCs were induced to differentiate with DMEM containing β-mercaptoethanol or Chinese angelica,respectively.Neuron-specific enolase(NSE),neurofilament(NF) were detected by immuohistochemistry.The results of the experiment indicate that BMSCs of goat in low abundance grow quickly when cultured in vitro ; BMSCs were differentiated into neurocytes,when treated with β-mercaptoethanol or Chinese angelica .The expression of NSE and NF in the neurocytes was positive.It is concluded that BMSCs of goat could be isolated,cultured in vitro ,and induced to differentiate into neurocytes with β-mercaptoethanol or Chinese angelica.
