摘要
利用RAPD技术,采用从80个随机引物中筛选的12个随机引物,对砀山地区梨树24个品种的基因组DNA进行扩增,通过3种编码方式(0 1编码,0 3编码,强带编码)对扩增的谱带进行标记并进行聚类分析。结果表明,运用特异性条带可鉴定梨树的品种类型、营养系变异和杂交种;白梨与沙梨具有较近的亲缘关系。在RAPD带型分析中,以0 3编码比0 1编码和强带编码反映的信息更全面。
The genomic DNA of 24 pear biotypes, which cultivated in Dangshan, Anhui Province, was amplified by the RAPD analysis by using the twelve arbitrary primers screened from 80 ones. The relationship between biotypes were studied by cluster analysis according to the RAPD patterns, which were coded in three ways: coding as 0-1, coding as 0-3 and the strong-band coding. The result showed that the cultivars, mutants and hybrids could be identified by using special bands. There was a close relationship between Pyrus bretschneideri and P.pyrifolia.The 0-3 coding could reflect more information than the other two coding ways.
