D-GalN大鼠模型肝卵圆细胞的活化和增殖

摘要 目的 探讨用D-氨基半乳糖(D-GalN)建立大鼠肝卵圆细胞(HOC)增殖模型。方法 予SD大鼠D-GalN 2.0g/kg体重腹腔注射后,分别于第1、2、3、4、5、6d处死大鼠(处死前1h腹腔注射BrdU 50 mg/kg体重),取肝组织分别行电镜、HE染色、免疫组化及RT-PCR检测。结果 电镜下可见HOC;HOC胞质OV-6染色阳性,胞核BrdU染色阳性;RT-PCR示模型组GGTmRNA表达增多。结论 用D-GalN 2.0 g/kg体重腹腔注射可以建立大鼠HOC增殖模型,但尚需优化。 Objective To establish a rat model with D-galactosamine (D-GalN). Methods Each male SD rat was administered with D-GalN intraperitoneally at the dose of 2.0 g/kg body weight. Animals in control group were received no treatment. Three rats from each group were killed every 24 h after the treatment. A single injection of 5' -bromodeoxyuridine(BrdU) was given intraperitoneally at the dose of 50 mg/kg body weight to the rats 1 h before sacrifice. Liver samples were taken for electron microscopy, immunohistochemistry and RT-PCR evaluation. Results Hepatic oval cells (HOC) were found under electron microscopy, and were characterized with brown staining in the cytoplasm for OV- 6 and black staining in the nuclei for anti-BrdU. RT-PCR showed the expression of GGTmRNA was much more than the controls. Conclusion A rat model for HOC proliferation can be established with D-GalN 2. 0 g/kg body weight.
