摘要
目的 研究焦点粘附激酶 (FAK )在肝细胞癌 (HCC)中的表达、活化及其临床意义。方法 采用原位分子杂交法检测 3 5例肝细胞癌 ,8例肝硬变及 5例正常肝组织中FAKmRNA的表达 ,并通过Western印迹杂交法分析肝癌组织中FAK的磷酸化 (Tyr -3 97)状态。 结果 肝癌组织中FAKmRNA水平明显高于肝硬变、正常肝组织 ( P <0 .0 1) ;同时低分化、存在转移的肝癌组织中FAK磷酸化水平明显高于高分化、无转移的肝癌 (P <0 .0 5 )。结论 FAK的高表达及磷酸化水平的上调可能参与了肝癌的发生发展 ,检测FAK的磷酸化 (Tyr -3 97)
Objective To investigate the expression and activation of focal adhesion kinase (FAK) in hepatocellular carcinoma (HCC),aswell as to elucidate its clinical significance.Methods FAK mRNA expression was detected by in situ hybridization in 35 cases of HCC,8 cases of liver cirrhosis and 5 cases of normal liver tissues;and the phosphorylations of FAK were analyzed by Western blotting.Results The expression levels of FAK mRNA in HCC were higher than those in liver cirrhosis and normal tissues(P<0.01).The phosphorylation of FAK was related to the differentiation degree of HCC and the status of metastasis(P<0.05).Conclusion The up-regulation of FAK mRNA and phosphorylation may contribute to carcinogenesis and progression of hepatocellular carcinoma.
