富马毒素B1诱导的小鼠肝细胞凋亡及端粒酶活性改变

摘要 目的 了解富马毒素 B1 ( FB1 )诱导小鼠肝细胞凋亡与端粒酶活性改变之间的关系。方法 给实验BALB/c小鼠喂饲含 1 mg/ml FB1饮用水 ,分别在不同时间取小鼠肝组织 ,用 TUNEL方法观察小鼠肝细胞凋亡发生情况 ,同时用 RT-PCR-ELISA法检测肝细胞端粒酶活性。结果 在摄入 FB1时间不同的各个实验组小鼠肝组织中均检测到端粒酶活性并观察到肝细胞凋亡明显增加。端粒酶活性随摄入 FB1时间增长而明显升高 ;但肝细胞凋亡与端粒酶活性变化有时相差异 ,表现为凋亡细胞指数先明显升高 ,然后逐渐减少的特点。结论FB1诱导小鼠肝细胞凋亡和其致癌作用是同时存在的 ,但表现为初期以细胞凋亡为主要损害形式 。 Objective To investigate the fumonisin B1(FB1)-induced apoptosis in connection with change in telomerase activity in liver cells of mice. Methods The BALB/c mice were given the water contained 1 mg/ml FB1 for 30 days, 60 days and 90 days respectively. At the end of experiment, the apoptotic cells were observed by TUNEL technic and the telomerase activity was measured with RT-PCR-ELISA method in liver cells of all experimental mice.Results After the mice were treated with FB1 for different periods, some classical morphological changes of apoptosis were observed under microscope and the telomerase activity increased in liver of all mice. But the apoptosis did not consist with the changes of telomerase activity. The telomerase activity increased constantly and the apoptotic cells appeared more in earlier phases, and then decreased gradually. Conclusion The effects of FB1-induced apoptosis and hepatocancerogenic activity are exist simultaneously. In earlier phases, FB1-induced apoptosis would be the main pathologic picture and FB1 might induce liver cancer later on.
