摘要
目的 研究腹腔注射 1 甲基 4 苯基 1,2 ,3,6 四氢吡啶 (MPTP)对褐鼠黑质纤连蛋白基因表达的影响 ,初步探讨其参与注射MPTP后的免疫机制。方法 通过腹腔注射MPTP建立帕金森病 (PD)褐鼠模型 ,采用逆转录 (RT) 聚合酶链反应 (PCR)方法研究纤连蛋白在黑质的表达情况。结果 经MPTP腹腔注射后褐鼠模型出现典型PD症状 ,黑质酪氨酸羟化酶表达水平明显降低 ,黑质纤连蛋白基因表达明显上调。结论 纤连蛋白在PD鼠黑质高表达可能促使T淋巴细胞透过血 脑屏障 (BBB)进入脑组织 ,参与PD鼠的免疫反应。
Objective To study the effects on gene expression of fibronectin in the substantia nigra of C57BL mice and the immune mechanism by peritoneal administration of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP).Methods The C57BL mouse models of Parkinson's disease(PD) were made by peritoneal administration of MPTP. Reverse transcriptase-polymerase chain reaction(RT-PCR) technique was used to detect the differential expression of fibronectin in the substantia nigra.Results The PD models were verified successfully by typical symptoms and obvious reduction of tyrosine hydroxylase expression in substantia nigra after peritoneal administration of MPTP.The result of RT-PCR revealed higher expression of fibronectin in the substantia nigra in PD mice.Conclusion Fibronectin may play an important role in immune mechanism of PD mice by promoting T-lymphocytes into brain tissue through blood brain barrier.
