MK-801与七叶皂苷纳联合应用对损伤脊髓细胞的保护作用 被引量:4

PROTECTIVE EFFECTS OF MK-801 COBINED WITH SODIUM AESCINATE THERAPY ON RATS WITH SPINAL CORD INJURY

摘要 目的探讨N-甲基-D-天门冬氨酸(NMDA)受体拮抗剂MK-801与七叶皂苷钠联合应用对损伤脊髓细胞的保护作用。方法60只大鼠随机分为治疗组、对照组和正常组,采用改良Allen背侧打击法、损伤脊髓。治疗组给予MK-801与七叶皂苷钠,对照组给生理盐水。受伤后1d、3d检测脊髓组织SOD活性及MDA食量,并用免疫组化方法检测脊髓组织内神经生长和修复相关GAP-43,MAP-2和CyclinD1的表达情况。结果对照组SOD活性降低,MDA含量增高(P<0.05),神经组织呈破坏性改变,损伤区周GAP-43、MAP-2和CyclinD1表达增高(P<0.05),治疗组可减轻脊髓组织损害,并增强GAP-43和MAP-2的表达(P<0.01),抑制CyclinD1的表达。结论脊髓损伤后MK-801与七叶皂苷钠联合应用可减轻脊髓组织水肿,清除自由基,抑制细胞凋亡和蛋白水解,对损伤脊髓细胞有明显保护作用,并促进神经组织的修复。 Objective: To study protective effects of the MK-801 combined with sodium aescinate therapy on rats with spinal cord injury. Methods: 60 rats were radomly divided into 3 group,treatment group(n=24),control group (n=24) and normal group (n=12). MK-801 combined with sodium aescina were used in the treatment group,while physiological saline was used in control group. The injury of spinal cord was made by proved Allen knock method. One or three days later,the rats were decapitated and their spinal cord were extracted for SOD activity and MDA concentration assay. Immunotistochemistry was used to evaluate the expression of GAP-43,MAP-2 and cyclin D. Results: SOD activity of control group decreased after spinal cord injurg while MDA contentration increased,compared with normal rats (P < 0.05) and the all levels of GAP-43,MAP-2 and cyclin D expressions were increased in injury areas (P < 0.05). The treatment group showed to reduce the injury of spinal cord and to enhance the expressions of GAP-43 and MAP-2 (P < 0.01), and to inhibit expression of cyclin D. Conclusion: MK-804 Combined with sodium aescinate therapy can prevent secondery injury of spinal cord by means of wiping free radical out, inhibiting apoptosi an promoting nerve regenaration.
