摘要
目的 :探讨葛根素对压力超负荷兔心力衰竭及其对心钠素 (ANP)和血管紧张素Ⅱ (AngⅡ )的影响。 方法 :将兔随机分为假手术组、手术对照组和葛根素组 ,采用腹主动脉次全结扎 ,建造兔心力衰竭模型 ;经颈动脉插管 ,利用BL -4 2 0E生物机能实验系统记录平均动脉压 (MAP)、心率 (HR)、左室收缩峰压 (LVSP)、左室舒张末压 (LVEDP)、最大上升速率 ( +dp/dtmax)和最大下降速率 ( -dp/dtmax)等血流动力学参数 ;放免法测定血浆ANP和AngⅡ水平。 结果 :( 1)手术对照组与假手术组相比 ,血流动力学参数明显异常 (P <0 .0 0 1) ;( 2 )葛根素组与手术对照组相比 ,HR减慢 ,±dp/dtmax明显上升 ,LVEDP明显下降 (P <0 .0 0 1) ;( 3 )葛根素组与手术对照组相比 ,血浆ANP浓度明显下降(P <0 .0 0 1) ,AngⅡ水平亦显著降低 (P <0 .0 5 )。 结论 :葛根素能减缓压力超负荷兔心力衰竭的发生 。
Objective:To evaluate the effect of puerarin on the rabbit with diastolic cardiac dysfunction and on the endocrine of atrial natriuretic peptide (ANP) and angiotensin Ⅱ (AngⅡ).Methods:30 rabbits were divided randomly into 3 groups:the sham operation group, the operation control group and the puerarin group. The rabbit models of diastolic heart faillure were made by the imcomplete ligation of abdominal aorta. Parameters of mean artery pressure (MAP), heart rate (HR), left ventricular systolic pressure (LVSP), left ventricular end-diastolic pressure (LVEDP) and the maximum rate of rise/decrease of left ventricular (±dp/dtmax) were measured through the tube inserted into the carotid artery and analysed by the BL-420E system software. Levels of plasma neurohormones, ANP and AngⅡ from the peripheral blood were measrued by radioactive immune assay (RIA) .Results:The rabbits of the operation group all occurred heart failure, while rabbits from puerarin group didn't occur heart failure.Heart rate and LVEDP decreased, ±dp/dtmax rised (P<0.001), levels of ANP decreased (P<0.001), and so did AngⅡ(P<0.05).Conclusions:The puerarin can attenuate or prevent the occurance of heart failure by decreasing the release of ANP and AngⅡ.
