摘要
目的 :探讨缝隙连接阻断剂甘珀酸对大鼠光化学局灶性脑梗死神经元损害的影响 .方法 :术前 2h右侧脑室注射甘珀酸 (5 0 μg/ 2 0 0g)使用光化学局灶性脑梗死模型 ,采用免疫组织化学和尼氏染色技术 ,观察甘珀酸预治疗对大鼠局灶性脑梗死 2 4h梗死灶体积、FOS及胶质原纤维酸性蛋白(GFAP)表达的影响 .结果 :光化学局灶性脑梗死后甘珀酸预治疗组梗死区体积较对照组明显减小 (P <0 .0 1 ) ;甘珀酸预治疗组大鼠脑梗死灶周边FOS表达较对照组明显减少 (P <0 .0 1 ) ;甘珀酸预治疗组大鼠脑梗死灶周边GFAP标记的星形胶质细胞发生了可塑性变化 ,但与对照组无差异 (P >0 .0 5 ) .结论
AIM: To explore the effects of carbenoxolone (CBX), a gap junction blocker, on the neuron injury induced by focal cerebral infarct. METHODS: Focal cerebral infarcts were produced by photothrombosis in adult rats. Immunohistochemical technique for anti FOS and anti GFAP, and Cresyl Violet staining were used in the present study. The volumes of focal cerebral infarct and the expressions of FOS and GFAP were studied 24 h after focal cerebral infarct. CBX(50 μg/200 g)was injected into the right lateral ventricle at 2h prior to the operation. RESULTS: The volumes of focal cerebral infarct and the expressions of FOS in the rats pretreated with CBX significantly decreased compared with those in the saline treated control group ( P <0.01). The GFAP positive astrocytes in both groups showed some changes in morphology, but with no significant difference between the two groups( P >0.05). CONCLUSION: The results suggest that CBX may play a role in protecting the neurons from damages induced by focal cerebral infarct.
